Cognition Therapeutics Accelerates Progress with Breakthroughs in Dementia with Lewy Bodies Treatment

Cognition Therapeutics has made significant strides in the treatment of dementia with Lewy bodies (DLB) through its commitment to advancing zervimesine, a unique small molecule designed to slow disease progression. The company recently announced alignment with the FDA on a planned registrational trial for zervimesine in individuals living with DLB, marking a major breakthrough in this area.

This achievement sets Cognition Therapeutics apart from other companies that have focused on symptomatic treatment of DLB through antipsychotics or medications for agitation. Zervimesine, however, is poised to make a meaningful impact by targeting the underlying disease process rather than just managing symptoms.

The company's President and Chief Executive Officer, Lisa Ricciardi, expressed pride in tackling an uncharted development path with the FDA, demonstrating Cognition Therapeutics' dedication to pushing the boundaries of what is possible. The proposed phase III trial will involve participants randomized to receive either 100 milligrams of zervimesine or a placebo orally once a day for nine months.

Dr. Tony Caggiano, Chief Medical Officer, highlighted the significance of zervimesine's unique properties, including its ability to bind to the sigma-2 receptor and protect neurons from toxic proteins without the need for brain shuttle technology. Additionally, the company has an extensive patent estate on zervimesine, with a recent USPTO patent issuance providing protection through 2045.

The proposed study schematic for the phase III trial builds upon the success of the phase II SHIMMER study, which demonstrated safety and tolerability in addition to showing meaningful impact across core features of DLB. The SHIMMER study evaluated neuropsychiatric symptoms using the NPI-12 tool, a measure that includes psychosis defined as hallucinations and delusions.

Cognition Therapeutics' commitment to advancing zervimesine represents a critical step forward in addressing the complex needs of individuals living with DLB. By prioritizing disease modification over symptomatic treatment, the company is poised to make a meaningful difference in the lives of those affected by this debilitating condition."
