Acumen Pharmaceuticals Steers Course Towards Breakthroughs in Alzheimer's Disease Treatment

Acumen Pharmaceuticals has made significant strides towards revolutionizing the treatment of Alzheimer's disease, as evident from its recent third-quarter 2026 conference call. The company's CEO, Dan O'Connell, and CFO, Matt Zuga, presented a detailed update on the progress made in their phase II ALTITUDE-AD trial investigating sabirnetug, their lead product candidate.

With expectations to announce top-line results this November, Acumen is poised to confirm its central scientific thesis that arresting harm done by synaptotoxic amyloid beta oligomers in the brain may offer a differentiated approach to treating Alzheimer's disease. The ALTITUDE-AD trial is a large and well-powered phase II study with clinically meaningful endpoints, testing two doses of sabirnetug against placebo.

Acumen has executed the study to an exceptionally high operational standard, with rapid enrollment, close engagement with study sites and investigators, and high rollover into the open-label extension. The company envisions this study as part of a potential registration package, with a single confirmatory phase III anticipated following positive phase II results.

The primary efficacy endpoint of the trial is the iADRS, which will be assessed following 18 months of treatment. Acumen expects the top-line data set to encompass primary endpoint results, key secondary outcomes, including the CDR-SB, safety evaluations covering adverse events and ARIA rates at a range of informative fluid and imaging biomarkers.

Acumen's product candidate, sabirnetug, has the potential to demonstrate a differentiated benefit-to-risk profile given its unique attributes as a selective anti-A-beta oligomer IgG2 monoclonal. The company is confident that the ALTITUDE-AD trial will provide valuable insights into the efficacy and safety of sabirnetug.

Acumen continues to advance its Enhanced Brain Delivery program, which represents the only program combining a validated blood-brain barrier penetrating technology with an A-beta oligomer selective therapeutic antibody. The company is working on two candidates in the lead-up to an IND filing expected in mid-2027, including ACU301 and ACU401.

The EBD bispecific antibodies, particularly ACU401, have shown encouraging results, achieving multifold greater brain exposure than unmodified ACU234 alone. The degree of brain penetration observed in non-human primates has exceeded expectations, giving confidence to the differentiated potential of Acumen's EBD program.

Acumen Pharmaceuticals' progress in Alzheimer's disease treatment marks a significant milestone towards a breakthrough. With its innovative approach and promising results, the company is well-positioned to make a meaningful impact on the lives of millions affected by this devastating disease.
